How to Make an Orthotopic Model of Oral Tongue Cancer in mouse?
(Protocol for an orthotopic oral tongue cancer model in mouse)
Materials:
1. 30 gauge needle
2. A 1-cc syringe
3. Q-tips
4. Tumor cells
Anesthetics:
Numbutal (pentobarbital sodium)
Methods:
1. Anesthetize a mouse by intraperitoneal injection with 60-90mg/kg body weight of pentobarbital sodium.
2. Open the mouse’s mouth and gently pull out the tongue using forceps with blunt teeth.
3. Insert 30-Gauge needle through the mucosa from the site near tongue root, advance the needle submucosally towards to the tongue tip, administer 50 ul of cell suspension.
4. Retract the needle.
5. Place a Q-tip over the injection site for about 30 second to prevent bleeding and spillage of material.
