How to make an orthotopic model of lung cancer (Intrathoracic injection) in mouse?

Materials:
1. 70% ethanol wipes and Betadine swabs.
2. Q-tips
3. 30-gauge needles
4. A 1-cc syringe
5. Tumor cells
Anesthetics:
Nembutal (pentobarbital sodium)
Methods:
The lung lies in the thoracic cavity. While the mouse left lung has only one single lobe, the right lung consists of cranial, middle, caudal and accessory lobes.
1. Anesthetize a mouse by intraperitoneal injection with 60-90mg/kg body weight of pentobarbital sodium.
2. Place it in the right lateral decubitus position.
3. Scrub the left lateral chest wall with Betadine followed by 70% alcohol.
4. Insert a 30-gauge needle attached to a 1-cc syringe slowly into the intercostal space at the dorsal mid-axillary line just below the inferior border of the scapula (about 1.5 cm above the lower rib line).
5. Advance the needle approximately 5 mm through the chest wall into the pleural space.
6. Inject 0.1 ml cell suspension.
7. Retract the needle.
8. Turn mouse to the left lateral decubitus position and keep the mouse warm to recover.
