How to Make an Orthotopic Model of Liver Cancer in mouse?
(Protocol for an orthotopic hepatic/liver cancer model in mouse)

Materials:

1. Surgical scissors, forceps, and scalpel.
2. 70% ethanol wipes and Betadine swabs.
3. An autoclip wound applier and stainless-steel autoclips.
4. Q-tips
5. 27- or 30-gauge needles
6. A 1-cc syringe
7. Tumor cells

Methoxyflurane

Methods:

The mouse liver is a large solid organ, lies in the right cranial part of the abdominal cavity, immediately dorsally to the diaphragm and right to stomach.

1. Anesthetize a nude mouse with methoxyflurane inhalation.
2. Place the mouse on its back.
3. Scrub the abdomen wall with Betadine followed by alcohol.
4. Incise the skin and peritoneum through the upper midline for about 1.5 cm. Expose the liver and stabilize its left lobe gently.
5. Insert a 27- or 30-gauge needle on a 1-cc syringe into the liver parenchyma just below the capsule in the left lobe and carefully advance the tip of needle along the capsule 3-4mm.
6. Inject the material slowly. A visible pale wheal indicates a successful injection. Retract the needle.
7. Place a Q-tip over the injection site for about 30 seconds to prevent bleeding and spillage of material.
8. Return the left lobe to the original place.
9. Close the wound with wound clip in one layer.
