How to Make an Orthotopic Model of Breast Cancer in Mouse?
(Protocol for an orthotopic breast cancer model in mouse)
Materials:
1. Surgical scissors, forceps, and scalpel.
2. 70% ethanol wipes and Betadine swabs.
3. An autoclip wound applier and stainless-steel autoclips.
4. Q-tips
5. 30-gauge needles
6. A 1-cc syringe
7. Tumor cells
Anesthetics:
Nembutal (sodium pentobarbital)
Methods:
A female mouse has five pairs of mammary glands, three pairs in the thoracic region and two in the inguino-abdominal region.
1. Anesthetize a mouse by intraperitoneal injection of Nembutal.
2. Restrain the mouse on the back.
3. Disinfect the skin over the lateral thorax with Betadine and 70% ethanol. Incise the skin just inferior to the nipple for about 5 mm long to reveal a mammary fatpad.
4. Insert a 30-gauge needle attached to a 1-cc syringe into the mammary fatpad.
5. Inject 50 µL of tumor cell suspension into the fatty tissue.
6. Retract the needle.
7. Close the skin with wound autoclip.
